Ovine pulmonary adenocarcinoma (OPA) is a naturally occurring contagious lung neoplasia caused by jaagsiekte sheep retrovirus (JSRV). Although no specific circulating antibodies against the virus can be detected in infected sheep, JSRV proviral DNA sequences can be found in peripheral blood leukocytes (PBLs) in clinically affected and in a proportion of in contact animals. In this study, existing hemi-nested PCR procedure is compared with a new one-step PCR technique that was developed to minimise potential DNA contamination and reduce sample and reagent handling. Different blood preparations were assessed and the best results were achieved on DNA prepared from buffy coat. The sensitivity of this PCR was lower in JSRV infected sheep without lesions of OPA than in clinically affected sheep, which indicate that this PCR may not be not fully appropriate for screening of individual sheep, but rather to provide results at flock level. This PCR is the only currently available blood test for detection of JSRV infected sheep and may be useful in epidemiological studies and in control programmes of OPA. 相似文献
The objective of this study was to examine the intralitter variation in postnatal growth performance, meat quality, and muscle fiber characteristics when littermates were categorized by carcass weight. Thirty-nine litters were weaned at 4 wk of age and had free access to feed from 2 wk of age until slaughter. They were slaughtered by litter at an average BW of 104 +/- 14 kg, and six pigs per litter were selected for analysis: the heaviest- (HW), middle- (MW), and lightest-weight (LW) pig of each sex. Categorizing littermates in LW, MW, and HW pigs at the same age reflected the differences in postnatal growth rate within a litter; thus ADG, muscle mass, and muscle deposition rate differed across pig weight groups (P < 0.001). Also, the total DNA content was different among pig weight groups (P < 0.001) and reflected differences in muscle growth rate. The difference in muscle growth rate between LW and MW pigs could be explained by a larger (P < 0.05) mean fiber area (MFA) in MW pigs, whereas the number of muscle fibers was similar. Growth rate differences between MW and HW pigs could in part be explained by a higher number (P < 0.01) of equal-sized muscle fibers in HW pigs. The difference in MFA was due to a higher estimated DNA and RNA content per muscle fiber in MW and HW compared with LW pigs (P < 0.05). Pigment content was higher in MW and HW compared with LW pigs (P < 0.01), but no other measured meat quality traits were significantly different across pig weight groups. These results indicate that both the number and the growth rate of muscle fibers contribute to intralitter variation in postnatal growth performance. 相似文献
This study was carried out to establish the changes in the free amino acid contents of floral honeys, honeydew honeys, and blend honeys during storage at room temperature and to test the capacity of the amino acids to distinguish the origin of the honeys after storage. For this purpose, 54 artisanal honeys (39 floral, 5 honeydew, and 10 blend) were studied. Samples were taken from recently collected honeys and at 3, 6, 9, 12, 16, 20, and 24 months after harvesting. The contents of most of the free amino acids were found to decrease with storage time, with the greatest reduction observed in the first 9 months. The contents of the amino acids aspartic acid, beta-alanine, and proline increased in the first few months after storage, reaching maximum values at 6 months, suggesting the possible existence of enzymatic activities. The application of stepwise discriminant analysis to the free amino acid content data demonstrated that the contents of the amino acids valine, beta-alanine, gamma-aminobutyric acid, serine, isoleucine, alpha-alanine, ornithine, and glutamine correctly assigned 87% of honeys to their group of origin: floral, honeydew, or blend. 相似文献
From the polar extracts of Origanum vulgare L. ssp. hirtum 19 compounds have been isolated. The structures and relative stereochemistry have been elucidated by spectroscopic analysis and determined as apigenin, luteolin, chrysoeriol, diosmetin, quercetin, eriodictyol, cosmoside, vicenin-2, caffeic acid, p-menth-3-ene-1,2-diol 1-O-beta-glucopyranoside, thymoquinol 2-O-beta-glucopyranoside, thymoquinol 5-O-beta-glucopyranoside, thymoquinol 2,5-O-beta-diglucopyranoside, 12-hydroxyjasmonic acid, 12-hydroxyjasmonic acid 12-O-beta-glucopyranoside, lithospermic acid B, rosmarinic acid, 10-epi-lithospermic acid, and epi-lithospermic acid B. The three latter products display unusual stereochemistry of the 3,4-hydroxyphenyllactic acid unit(s), which to the authors' best knowledge has never been reported before in similar compounds. Moreover, lithospermic acid B (and its stereoisomers), p-menth-3-ene-1,2-diol 1-O-beta-glucopyranoside, 12-hydroxyjasmonic acid, and 12-hydroxyjasmonic acid 12-O-beta-glucopyranoside were isolated for the first time from Origanum species. 相似文献
Abstract 1. Carcass cuts, chemical composition, colour, textural parameters, fatty acid and amino acid profiles, macro- and micromineral content and sensory properties of breast and drumstick meat from 10 month old common pheasant produced in an extensive rearing system were analysed.
2. Breast muscle was characterised by a high concentration of protein (254 g/kg), low content of intramuscular fat (1.3 g/kg), considerable amount of Fe-haeme (4.9 mg/kg) low shear force (1.96 kg/cm2) and hardness (4.07 kg/cm2).
3. The main drumstick characteristics were 222 g of protein/kg, 4.0 g of intramuscular fat/g and a significant higher cholesterol concentration compared to that of breast (7.7 vs. 2.9 g/kg).
4. Saturated, monounsaturated and polyunsaturated fatty acid mean values for both muscle types were approximately 34.5%, 43.3% and 24.2% of total fatty acid methyl esters, respectively, and n-3 content in breast was significantly higher (3.21% vs. 1.65%). Significant differences were found between the three tissues (breast, drumstick and subcutaneous fat).
5. Large differences in the amino acid profile between breast and drumstick were detected, especially in the non-essential fraction. The main essential amino acids were lysine and leucine, whereas aspartic and glutamic acids were the most important non-essential amino acids.
6. There were significant differences between muscle types in the mineral content with regard to iron, magnesium, sodium and zinc. Pheasant meat is a good nutritional source of iron as 100 g of drumstick may provide 23.6% of the recommended daily amount. 相似文献
The endemic non-pathogenic Australian rabbit calicivirus RCV-A1 is known to provide some cross protection to lethal infection with the closely related Rabbit Haemorrhagic Disease Virus (RHDV). Despite its obvious negative impacts on viral biocontrol of introduced European rabbits in Australia, little is known about the extent and mechanisms of this cross protection. In this study 46 rabbits from a colony naturally infected with RCV-A1 were exposed to RHDV. Survival rates and survival times did not correlate with titres of serum antibodies specific to RCV-A1 or cross reacting to RHDV, but were instead influenced by the time between infection with the two viruses, demonstrating for the first time that the cross protection to lethal RHDV infection is transient. These findings are an important step towards a better understanding of the complex interactions of co-occurring pathogenic and non-pathogenic lagoviruses. 相似文献
This study investigated the responses of soil animal communities, soil functioning and humus forms to forest dynamics and solar radiation. We examined changes in invertebrate communities and soil features in two subalpine spruce forests (Eastern Italian Alps, Trento) growing on a calcareous bedrock, with different sun exposures (north and south), each forming a chronosequence of three developmental phases: clearing, regeneration stand (25-year-old trees) and mature stand (170-year-old trees). Our results indicate that the two forest sites differed in solar energy input, soil chemical properties and the relationships between forest dynamics and animal communities. In the north-facing site, soil fauna communities were very similar in the three forest developmental phases. Conversely, in the south-facing site, the composition of invertebrate communities and the diversity of zoological groups varied greatly among developmental phases. The highest abundance of total invertebrates, and mites in particular, occurred in the south-facing mature stands while the south-facing regeneration stand was characterised by higher densities of Collembola, Chilopoda, Symphyla, Protura and Aranea. The structure of communities in clearings was the same as in regeneration stands but with lower invertebrate abundance. Humus forms and soil features changed with developmental phases in both the south- and north-facing sites, although variations were more pronounced in the southern exposure. Mature stands were characterised by high levels of soil organic carbon and nitrogen, C/N values and low pH, the clearings and regeneration stands being characterised by a greater release of mineral nitrogen. The diversity of zoological groups (Shannon–Wiener index) was linearly correlated to soil pH, Humus Index, the amount of organic carbon and the species richness of herbaceous plants. Our results about the composition and the diversity of invertebrate communities are consistent with the observations of other authors studying south-exposed forests growing on different bedrock types, indicating that such relationships are widespread. The higher densities of invertebrates in the south-facing site may be attributed to higher solar radiation, and the positive correlation observed between total soil fauna abundance and solar energy supports the “more individuals” hypothesis that assumes a positive relationship between the number of individuals and energy availability. Possible ways by which forest dynamics control soil invertebrate communities are discussed. 相似文献
